唐木英明・東京大名誉教授（毒性学・薬理学）は、プベルル酸について詳細は不明だとしたうえで、「青カビが作る物質であるなら、紅こうじ菌から自然に作られることはありえない」と指摘。製造過程で青カビが混入した可能性があるが、紅こうじを作る培養器はカビも非常に増えやすい環境のため、混入すれば一気に増殖し、色の違いで一目で分かるはずだという。
また、プベルル酸が腎臓にどのような影響を及ぼすのかも明らかになっていない。「腎臓への毒性があるかを動物実験で調べることが必要だが、結果が出るまで1～2カ月はかかるのではないか」とみる。
医薬品の製造現場に詳しい光本泰秀・北陸大教授（神経薬理学）もプベルル酸について、「なぜ腎臓に集積したのかや、もしくは集積していなくても、腎臓に毒性を示したのかを明らかにしなければ、原因物質として特定できない」と指摘する。そのうえで「これまで紅こうじを使った製品を長年製造してきたなかで、急に特定のロットの製品で健康被害が出るのは不思議だ。外部からの混入などの可能性も含めて調査する必要がある」と話した。

However, since the culture vessels used to produce red yeast rice are environments where mold can easily proliferate, any contamination should rapidly multiply and be easily recognizable by differences in color.Furthermore, the effects of citrinin on the kidneys are still unknown. Professor Yasuhide Mitsumoto, an expert in pharmaceutical manufacturing processes at Hokuriku University (Neuropharmacology), also emphasized that "without clarifying why citrinin accumulated in the kidneys or whether it showed toxicity to the kidneys even without accumulation, it cannot be identified as the causative substance." He added, "It's strange that health issues suddenly arose in specific batches of products after years of manufacturing products using red yeast rice. Investigation is needed, including the possibility of external contamination."
